A Microfluidic Device to Mimic the Bone Marrow Microenvironment: Real-time Observation of the Leukemic Cell Behavior
نویسندگان
چکیده
We investigated the possibility of mimicking the bone marrow (BM) microenvironment using a microfluidic device. We succeeded in observing the differentiation into osteoclasts (OCs) and the migration of leukemic cells into the mimicked BM microenvironment. We confirmed that the BM microenvironment was mimicked in vitro successfully. These findings suggest that this attractive device can contribute to medical research.

Alterations of adiponectin gene expression in bone marrow of acute myeloid leukemia
Background: Acute myeloid leukemia (AML) is characterized by the proliferation of myeloid precursors and abnormal differentiation of hematopoietic stem cells, which results in the accumulation of immature cells in the bone marrow (BM). The accumulation of these cells in the bone marrow causes molecular and cellular changes in the microenvironment of the bone marrow. The adiponectin hormone orig...

Hematopoietic stem/progenitor cells (HSPCs) which give rise to different blood cell types are present within the bone marrow microenvironment, especially in flat bones such as skull, vertebrae, pelvis and chest. Interacting factors such as stromal derived factor-1/CXCR4, very late antigen-4/vascular cell adhesion molecule-1, Lymphocyte function-associated antigen-1/ intercellular adhesion molec...
متن کاملThree-Dimensional Microfluidic Tri-Culture Model of the Bone Marrow Microenvironment for Study of Acute Lymphoblastic Leukemia
Acute lymphoblastic leukemia (ALL) initiates and progresses in the bone marrow, and as such, the marrow microenvironment is a critical regulatory component in development of this cancer. However, ALL studies were conducted mainly on flat plastic substrates, which do not recapitulate the characteristics of marrow microenvironments. To study ALL in a model of in vivo relevance, we have engineered...
متن کاملBone Tissue Engineering: a Mini-Review
Despite advances in bone tissue engineering, auto grafts from intra-oral or extra-oral donor sites are still the gold standard for treatment of large craniomaxillofacial defects. Biomaterial development, application of growth factor, and stem cells, open new gateway to bone regeneration studies, but real translation from bench to bedside have not yet happened. In this review article, a number o...
متن کاملLeukemic cells create bone marrow niches that disrupt the behavior of normal hematopoietic progenitor cells.
The host tissue microenvironment influences malignant cell proliferation and metastasis, but little is known about how tumor-induced changes in the microenvironment affect benign cellular ecosystems. Applying dynamic in vivo imaging to a mouse model, we show that leukemic cell growth disrupts normal hematopoietic progenitor cell (HPC) bone marrow niches and creates abnormal microenvironments th...
